WHAT IS GRISEOFULVINE?

Griseofulvin is an old drug used to treat fungal infections. Before modern antifungal tablets (azoles) came on the market, griseofulvin was the only available medication. Because the newer medications had fewer side effects than griseofulvin, it was discontinued in the Netherlands for a while. However, it turned out that griseofulvin is still occasionally necessary for certain types of fungal infections, particularly scalp fungus. Certain types of fungi don't respond as well to the newer medications, but they do to griseofulvin.
Griseofulvin needs to be used for a longer period than the other medications and has more side effects, but it is effective.

Other medications, such as terbinafine and itraconazole, are used to treat nail fungus.

IS GRISEOFULVINE AVAILABLE WITHOUT A PRESCRIPTION?

Griseofulvin is a prescription-only medication. This means it is only available with a doctor's prescription.

WHEN CAN I USE GRISEOFULVINE?

Applications (= indications): e.g.
Infections with fungi of skin, hair and nails

WHEN NOT TO USE?

Do not use in case of (= contraindications)
- Liver function disorders
- Lupus erythematosus (autoimmune disease)
- Hypersensitivity to this medicine
- Porphyria (= disorder in the formation of red blood pigment)

PREGNANCY AND BREASTFEEDING

Do not use this medication during pregnancy. Griseofulvin can cause birth defects in the unborn child.
Do not use this medication while breastfeeding.

TRAFFIC WORK AND SPORTS

This medicine does not affect your ability to react and concentrate during normal daily activities, work, sports or participation in traffic.

OPERATION

This is an antifungal agent (= antimycotic) that is effective against fungi of the skin, hair and nails that are sensitive to griseofulvin.

SIDE EFFECTS

- Allergic reactions, including skin rash (urticaria) and photosensitivity
- Vomiting
- Abdominal pain
- Depression
- Diarrhea
- Dry mouth
- Dizziness
- Headache
- Gastrointestinal complaints
- Nausea
- Fatigue
- Irritability
- Drowsiness
- Confusion
- Impaired liver function

INTERACTIONS

Potential interactions
: - Alcohol, which increases the effect of alcohol
- Barbiturates (anti-epileptic drugs)
- Anticoagulants (coumarin-type oral anticoagulants)
- Contraceptives (the pill). Concomitant use of griseofulvin may reduce the effectiveness of the contraceptive pill, potentially leading to an unwanted pregnancy.

INSTRUCTIONS FOR USE

Take this medicine with fatty food or with milk.

DOSAGE

Adults: 500 mg per day (1000 mg for severe infections) in 1-4 divided doses, for 6-8 weeks, longer if necessary.

Children: 10-20 mg/kg/day.
< 1 year (< 10 kg): 125 mg per day, e.g. 5 ml (= 125 mg) once daily, or 1 125 mg tablet once daily.
1-3 years (10-15 kg): 200-300 mg per day, e.g. 10 ml (= 250 mg) once daily, or 2 125 mg tablet once daily.
3-6 years (15-20 kg): 300-400 mg per day, e.g. 15 ml (= 375 mg) once daily, or 3 125 mg tablet once daily.
6-10 years (20-30 kg): 400-500 mg per day, e.g. 15-20 ml (= 375-500 mg) once daily, or 3-4 tablets of 125 mg once daily.
> 10 years (> 30 kg): 500 mg per day, e.g. 4 tablets of 125 mg once daily or 2 tablets of 125 mg twice daily.
The dose can be taken at once. In case of nausea, divide it into 2 or more doses.
